چکیده

Introduction Dynamic contrast enhanced MRI (DCE-MRI) is used to characterize perfusion and extracellular volume in tumors through the tracer kinetic modeling of MRI contrast agents (typically Gd-DTPA). However, the parametric estimates resultant of DCE-MRI may be influenced by several confounding factors, including inter-compartmental water exchange [1]. In contrast to MRI contrast agents, radiotracer imaging with SPECT uses direct measures of tracer activity and is, therefore, insensitive to water dynamics. Here we compare extracellular volume, ve, in a C6 glioma model in rat brain as measured by DCE-MRI and dual-isotope SPECT.

Can DCE-MRI Explain the Heterogeneity in Radiopeptide Uptake Imaged by SPECT in a Pancreatic Neuroendocrine Tumor Model?

Although efficient delivery and distribution of treatment agents over the whole tumor is essential for successful tumor treatment, the distribution of most of these agents cannot be visualized. However, with single-photon emission computed tomography (SPECT), both delivery and uptake of radiolabeled peptides can be visualized in a neuroendocrine tumor model overexpressing somatostatin receptors...

Glioma Grading Capability: Comparisons among Parameters from Dynamic Contrast-Enhanced MRI and ADC Value on DWI

OBJECTIVE Permeability parameters from dynamic contrast-enhanced MRI (DCE-MRI) and apparent diffusion coefficient (ADC) value on diffusion-weighted imaging (DWI) can be quantitative physiologic metrics for gliomas. The transfer constant (K(trans)) has shown efficacy in grading gliomas. Volume fraction of extravascular extracellular space (ve) has been underutilized to grade gliomas. The purpose...

Imaging heterogeneity of peptide delivery and binding in solid tumors using SPECT imaging and MRI

BACKGROUND As model system, a solid-tumor patient-derived xenograft (PDX) model characterized by high peptide receptor expression and histological tissue homogeneity was used to study radiopeptide targeting. In this solid-tumor model, high tumor uptake of targeting peptides was expected. However, in vivo SPECT images showed substantial heterogeneous radioactivity accumulation despite homogenous...

Measurement of rat brain tumor kinetics using an intravascular MR contrast agent and DCE-MRI nested model selection.

PURPOSE Using dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) in a rat glioma model, and nested model selection (NMS), to compare estimates of the pharmacokinetic parameters vp , K(trans) , and ve for two different contrast agents (CAs)-gadofosveset, which reversibly binds to human serum albumin, and gadopentetate dimeglumine, which does not. MATERIALS AND METHODS DCE-MRI studi...
